To sum up his answer:
First he says there are a few ideas frequently bandied about regarding his involvement in Broadway. One is a pastiche of his catalogue put together a la Tomfoolery (or Movin' Out or American Idiot). Another would be his standard touring show, but amped up for a run on Broadway. The third idea would be to write his own original Broadway musical which he says he is currently investigating, but doesn't want to talk about it much because these things tend not to pan out.
The host then follows up by asking if Al is a fan of Book of Mormon. Al says he is a fan of that show, a fan of South Park, but he can't talk too much about it because it may involve a project that is "in development" and closes by strongly insinuating that he "may or may not be working with" Matt & Trey.

The thing to remember with musicals on Broadway is that generally they are extraordinarily expensive to produce and a very high risk investment, especially new musicals. I think Tomfoolery was off-Broadway, which is less expensive. Yes, I would love to see a live theatrical piece stemming from Al. Don't know if it will actually take the Broadway route, though.
